Design Notes
mNG was chosen as the fluorescent protein, as it is the brightest available fluorescent protein. mNG was linked to the C-terminal, rather than the N-terminal, to avoid blocking the antigen binding site of the antibody. OmpA recognition protein sequence was placed upstream of the ScFv coding sequence to direct the protein to the periplasm.

The antibody fragment was produced by Dr. Brennan's "The Production of Recombinant single chain Antibody Fragments for the Detection of Illicit Drug Residues". The mNG sequence was obtained from Branchiostoma lanceolatum by Shaner et al. (2013).
